Semen is the fluid that (sparing people with conditions that change this for them) contains sperm cells and is released with a full ejaculation.
Pre-ejaculate (or "pre-cum") is a fluid that is usually also secreted by the penis, but often long before an ejaculation. All by itself it doesn't contain semen, and is though to occur in order to clear the urethra of old trace sperm cells and anything else so that if and when ejaculation occurs, it's all clear, basically.
